Abstract
The promoter and leader sequences (-2271-+178) of a soybean seed coat-specific gene SCSI (Seed Coat Subtilisin 1) were analyzed for regulatory activity by transient and stable expression in tobacco using GUS gene fusions in different vectors. The results showed that the region of -2271-+178 conferred seed coat-specific GUS expression in transgenic tobacco. The untranslated leader sequence (+1-+178) was shown to be responsible for the seed coat-specific activity, while the up stream region (-2271—50) was observed to control the quantity of expression. Enhanced GUS activity promoted by the leader sequence of the SCSI gene was also observed. In contrast, identical configuration of the SCSI promoter conferred high levels of transient GUS activity in leaf tissue when delivered into tobacco leaf cells by particle bombardment. In addition, it was observed that the presence of 35S promoter sequences in an expression vector could affect the expression pattern from promoters inserted into that vector.
